Increased Nitration of Sarcoplasmic Reticulum Ca -ATPase in Human Heart Failure

چکیده
Background—Reduced sarcoplasmic reticulum (SR) Ca -ATPase (SERCA2a isoform) activity is a major determinant of reduced contractility in heart failure. Ca -ATPase inactivation can occur through SERCA2a nitration. We therefore investigated the role of SERCA2a nitration in heart failure. Methods and Results—We measured SERCA2a levels and nitrotyrosine levels in tissue from normal and failing human hearts using Western blots. We found that nitrotyrosine levels in idiopathic dilated cardiomyopathic (DCM) hearts were almost double those of control hearts in age-matched groups. Nitrotyrosine was dominantly present in a single protein with the molecular weight of SERCA2a, and immunoprecipitation confirmed that the protein recognized by the nitrotyrosine antibody was SERCA2a. There was a positive correlation between the time to half relaxation and the nitrotyrosine/SERCA2a content (P 0.01) in myocytes isolated from control and DCM hearts. In experiments with isolated SR vesicles from porcine hearts, we also showed that the Ca pump is inactivated by peroxynitrite exposure, and inactivation was prevented by protein kinase A pretreatment. Conclusions—We conclude that SERCA2a inactivation by nitration may contribute to Ca pump failure and hence heart failure in DCM. (Circulation. 2005;111:988-995.)

متن کاملThe Effect of Verapamil Administred before the Reperfusion Insult in Isolated Preconditioned Rat Heart on the Microsomal ATPase and Mitochondrial Enzyme Activities
Background: Calcium overload and free radical mediated damage in the mitochondria is the most important pathological changes associated with myocardial ischemic-reperfusion injury. The verapamil post-treatment has been previously reported to prevent reperfusion-induced myocardial injury but functional recovery may be delayed due to the drug's inherent direct myocardial depression effect. In the...
متن کاملTargeted Overexpression of the Sarcoplasmic Reticulum Ca-ATPase Increases Cardiac Contractility in Transgenic Mouse Hearts
Cardiac hypertrophy and heart failure are known to be associated with a reduction in Ca-ATPase pump levels of the sarcoplasmic reticulum (SR). To determine whether, and to what extent, alterations in Ca pump numbers can affect contraction and relaxation parameters of the heart, we have overexpressed the cardiac SR Ca-ATPase specifically in the mouse heart using the a-myosin heavy chain promoter...
متن کاملS100A1 in human heart failure: lack of recovery following left ventricular assist device support.
BACKGROUND We hypothesized that S100A1 is regulated during human hypertrophy and heart failure and that it may be implicated in remodeling after left ventricular assist device. S100A1 is decreased in animal and human heart failure, and restoration produces functional recovery in animal models and in failing human myocytes. With the potential for gene therapy, it is important to carefully explor...
متن کاملCa(2+)-transporting ATPase, phospholamban, and calsequestrin levels in nonfailing and failing human myocardium.
BACKGROUND Observations of abnormalities in the diastolic components of intracellular Ca2+ transients in failing human left ventricular myocardium have raised the possibility that reductions in the level or function of sarcoplasmic reticulum proteins involved in Ca2+ transport contribute to the pathophysiology of dilated cardiomyopathy in humans. Functional assays, however, have revealed no dif...
